Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_003072.5(SMARCA4):c.2542C>G (p.Leu848Val), citing Ambry Variant Classification Scheme 2023: The p.L848V variant (also known as c.2542C>G), located in coding exon 17 of the SMARCA4 gene, results from a C to G substitution at nucleotide position 2542. The leucine at codon 848 is replaced by valine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
