Uncertain significance — the classification assigned by Ambry Genetics to NM_001377540.1(SLMAP):c.1657A>C (p.Asn553His), citing Ambry Variant Classification Scheme 2023: The p.N519H variant (also known as c.1555A>C), located in coding exon 16 of the SLMAP gene, results from an A to C substitution at nucleotide position 1555. The asparagine at codon 519 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
