NM_017841.4(SDHAF2):c.410del (p.Met137fs) was classified as Likely pathogenic for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.410delT variant, located in coding exon 4 of the SDHAF2 gene, results from a deletion of one nucleotide at nucleotide position 410, causing a translational frameshift with a predicted alternate stop codon (p.M137Rfs*4). This alteration occurs at the 3' terminus of theSDHAF2 gene, is not expected to trigger nonsense-mediated mRNA decay, and only impacts the last 18% of the protein. However, premature stop codons are typically deleterious in nature and the impacted region is critical for protein function (Ambry internal data). This variant has been observed in at least one individual with a personal and/or family history that is consistent with SDHAF2-associated disease (Ambry internal data). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). Based on the majority of available evidence to date, this variant is likely to be pathogenic.
