Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001283009.2(RTEL1):c.2450G>C (p.Cys817Ser), citing Ambry Variant Classification Scheme 2023: The p.C817S variant (also known as c.2450G>C), located in coding exon 26 of the RTEL1 gene, results from a G to C substitution at nucleotide position 2450. The cysteine at codon 817 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
