Uncertain significance — the classification assigned by Ambry Genetics to NM_021930.6(RINT1):c.302A>G (p.Lys101Arg), citing Ambry Variant Classification Scheme 2023: The p.K101R variant (also known as c.302A>G), located in coding exon 4 of the RINT1 gene, results from an A to G substitution at nucleotide position 302. The lysine at codon 101 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
