Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_020975.6(RET):c.1544G>T (p.Cys515Phe), citing Ambry Variant Classification Scheme 2023: The p.C515F variant (also known as c.1544G>T), located in coding exon 8 of the RET gene, results from a G to T substitution at nucleotide position 1544. The cysteine at codon 515 is replaced by phenylalanine, an amino acid with highly dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
