NM_001134363.3(RBM20):c.2141_2145dup (p.Arg716fs) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.2141_2145dupACCCC variant, located in coding exon 9 of the RBM20 gene, results from a duplication of ACCCC at nucleotide position 2141, causing a translational frameshift with a predicted alternate stop codon (p.R716Tfs*59). This alteration is expected to result in protein truncation or nonsense-mediated mRNA decay. However, loss of function of RBM20 has not been established as a mechanism of disease. Based on the available evidence, the clinical significance of this alteration remains unclear.
