Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_002875.5(RAD51):c.496T>G (p.Phe166Val), citing Ambry Variant Classification Scheme 2023: The p.F166V variant (also known as c.496T>G), located in coding exon 5 of the RAD51 gene, results from a T to G substitution at nucleotide position 496. The phenylalanine at codon 166 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
