Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005732.4(RAD50):c.1359G>T (p.Lys453Asn), citing Ambry Variant Classification Scheme 2023: The p.K453N variant (also known as c.1359G>T), located in coding exon 9 of the RAD50 gene, results from a G to T substitution at nucleotide position 1359. The lysine at codon 453 is replaced by asparagine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
