Uncertain significance for Nephrolithiasis/nephrocalcinosis — the classification assigned by Ambry Genetics to NM_000388.4(CASR):c.1008G>C (p.Lys336Asn), citing Ambry Variant Classification Scheme 2023: The p.K336N variant (also known as c.1008G>C), located in coding exon 3 of the CASR gene, results from a G to C substitution at nucleotide position 1008. The lysine at codon 336 is replaced by asparagine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
