NM_000314.8(PTEN):c.669G>C (p.Lys223Asn) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K223N variant (also known as c.669G>C), located in coding exon 7 of the PTEN gene, results from a G to C substitution at nucleotide position 669. In a massively parallel functional assay using a humanized yeast model, lipid phosphatase activity for this variant was functionally neutral (Mighell TL et al. Am J Hum Genet, 2018 May;102:943-955). The lysine at codon 223 is replaced by asparagine, an amino acid with similar properties. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
