Uncertain significance — the classification assigned by Ambry Genetics to NM_006904.7(PRKDC):c.388C>A (p.Leu130Ile), citing Ambry Variant Classification Scheme 2023: The p.L130I variant (also known as c.388C>A), located in coding exon 4 of the PRKDC gene, results from a C to A substitution at nucleotide position 388. The leucine at codon 130 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
