Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.1019A>G (p.Glu340Gly), citing Ambry Variant Classification Scheme 2023: The p.E340G variant (also known as c.1019A>G), located in coding exon 10 of the POLE gene, results from an A to G substitution at nucleotide position 1019. The glutamic acid at codon 340 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
