NM_006231.4(POLE):c.1469A>G (p.Asp490Gly) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D490G variant (also known as c.1469A>G), located in coding exon 14 of the POLE gene, results from an A to G substitution at nucleotide position 1469. The aspartic acid at codon 490 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
