NM_002691.4(POLD1):c.2257T>C (p.Tyr753His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y753H variant (also known as c.2257T>C), located in coding exon 18 of the POLD1 gene, results from a T to C substitution at nucleotide position 2257. The tyrosine at codon 753 is replaced by histidine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
