Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003661.4(APOL1):c.137A>G (p.Asp46Gly), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces aspartic acid, which is acidic and polar, with glycine, which is neutral and non-polar, at codon 46 of the APOL1 protein (p.Asp46Gly). This variant is present in population databases (rs144963144, gnomAD 0.03%). This variant has not been reported in the literature in individuals affected with APOL1-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant¬†is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
