Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006206.6(PDGFRA):c.434C>G (p.Ser145Cys), citing Ambry Variant Classification Scheme 2023: The p.S145C variant (also known as c.434C>G), located in coding exon 3 of the PDGFRA gene, results from a C to G substitution at nucleotide position 434. The serine at codon 145 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
