NM_006206.6(PDGFRA):c.1001A>G (p.His334Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H334R variant (also known as c.1001A>G), located in coding exon 6 of the PDGFRA gene, results from an A to G substitution at nucleotide position 1001. The histidine at codon 334 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
