Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000038.6(APC):c.1151G>C (p.Ser384Thr), citing Ambry Variant Classification Scheme 2023: The p.S384T variant (also known as c.1151G>C), located in coding exon 9 of the APC gene, results from a G to C substitution at nucleotide position 1151. The serine at codon 384 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, in silico predictors for this gene do not accurately predict pathogenicity. Based on the available evidence, the clinical significance of this variant remains unclear.
