Uncertain significance — the classification assigned by Ambry Genetics to NM_001386125.1(OBSCN):c.10341G>C (p.Glu3447Asp), citing Ambry Variant Classification Scheme 2023: The p.E3018D variant (also known as c.9054G>C), located in coding exon 34 of the OBSCN gene, results from a G to C substitution at nucleotide position 9054. The glutamic acid at codon 3018 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
