Uncertain significance — the classification assigned by Ambry Genetics to NM_001386125.1(OBSCN):c.19240C>G (p.Gln6414Glu), citing Ambry Variant Classification Scheme 2023: The p.Q5457E variant (also known as c.16369C>G), located in coding exon 62 of the OBSCN gene, results from a C to G substitution at nucleotide position 16369. The glutamine at codon 5457 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
