Uncertain significance — the classification assigned by Ambry Genetics to NM_002519.3(NPAT):c.374A>T (p.Lys125Met), citing Ambry Variant Classification Scheme 2023: The p.K125M variant (also known as c.374A>T), located in coding exon 6 of the NPAT gene, results from an A to T substitution at nucleotide position 374. The lysine at codon 125 is replaced by methionine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
