Uncertain significance — the classification assigned by Ambry Genetics to NM_002519.3(NPAT):c.475C>G (p.Arg159Gly), citing Ambry Variant Classification Scheme 2023: The p.R159G variant (also known as c.475C>G), located in coding exon 6 of the NPAT gene, results from a C to G substitution at nucleotide position 475. The arginine at codon 159 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
