Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_017617.5(NOTCH1):c.6359G>A (p.Arg2120His), citing Ambry Variant Classification Scheme 2023: The p.R2120H variant (also known as c.6359G>A), located in coding exon 34 of the NOTCH1 gene, results from a G to A substitution at nucleotide position 6359. The arginine at codon 2120 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
