NM_000268.4(NF2):c.931A>T (p.Arg311Trp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R311W variant (also known as c.931A>T), located in coding exon 10 of the NF2 gene, results from an A to T substitution at nucleotide position 931. The arginine at codon 311 is replaced by tryptophan, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
