NM_000268.4(NF2):c.806A>G (p.Lys269Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K269R variant (also known as c.806A>G), located in coding exon 8 of the NF2 gene, results from an A to G substitution at nucleotide position 806. The lysine at codon 269 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
