NM_001042492.3(NF1):c.89C>T (p.Thr30Ile) was classified as Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T30I variant (also known as c.89C>T), located in coding exon 2 of the NF1 gene, results from a C to T substitution at nucleotide position 89. The threonine at codon 30 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
