NM_001042492.3(NF1):c.7163T>G (p.Phe2388Cys) was classified as Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.F2367C variant (also known as c.7100T>G), located in coding exon 47 of the NF1 gene, results from a T to G substitution at nucleotide position 7100. The phenylalanine at codon 2367 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
