Uncertain significance — the classification assigned by Ambry Genetics to NM_006393.3(NEBL):c.1985A>G (p.Tyr662Cys), citing Ambry Variant Classification Scheme 2023: The p.Y662C variant (also known as c.1985A>G), located in coding exon 20 of the NEBL gene, results from an A to G substitution at nucleotide position 1985. The tyrosine at codon 662 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
