Uncertain significance — the classification assigned by Ambry Genetics to NM_003803.4(MYOM1):c.43C>A (p.Leu15Ile), citing Ambry Variant Classification Scheme 2023: The p.L15I variant (also known as c.43C>A), located in coding exon 1 of the MYOM1 gene, results from a C to A substitution at nucleotide position 43. The leucine at codon 15 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
