This variant lies in the MYLK gene (transcript NM_053025.4) at coding-DNA position 5715 through coding-DNA position 5720, duplicating 6 bases. Submitter rationale: The c.5715_5720dupTGAAGG variant (also known as p.G1909_E1910dup), located in coding exon 31 of the MYLK gene, results from an in-frame duplication of TGAAGG at nucleotide positions 5715 to 5720. This results in the duplication of 2 extra residues (GE) between codons 1910 and 1911. This amino acid region is not well conserved in available vertebrate species. In addition, this alteration is predicted to be neutral by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
