Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000071.3(CBS):c.1194GGA[2] (p.Glu400del), citing Ambry Variant Classification Scheme 2023: The c.1200_1202delGGA variant (also known as p.E400del) is located in coding exon 11 of the CBS gene. This variant results from an in-frame GGA deletion at nucleotide positions 1200 to 1202. This results in the in-frame deletion of a glutamic acid at codon 400. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be neutral by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
