Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000179.3(MSH6):c.3331A>G (p.Asn1111Asp), citing Ambry Variant Classification Scheme 2023: The p.N1111D variant (also known as c.3331A>G), located in coding exon 5 of the MSH6 gene, results from an A to G substitution at nucleotide position 3331. The asparagine at codon 1111 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
