This variant lies in the MSH2 gene (transcript NM_000251.3) at coding-DNA position 130, where A is replaced by T; at the protein level this means replaces threonine at residue 44 with serine — a missense variant. Submitter rationale: The p.T44S variant (also known as c.130A>T), located in coding exon 1 of the MSH2 gene, results from an A to T substitution at nucleotide position 130. The threonine at codon 44 is replaced by serine, an amino acid with similar properties. In a massively parallel cell-based functional assay testing susceptibility to a DNA damaging agent, 6-thioguanine (6-TG), this variant was determined to be functionally neutral (Jia X et al.
