Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000251.3(MSH2):c.2458+5del, citing Ambry Variant Classification Scheme 2023. This variant lies in the MSH2 gene (transcript NM_000251.3) at 5 bases into the intron immediately after coding-DNA position 2458, deleting one base. Submitter rationale: The c.2458+5delG intronic variant, located in intron 14 of the MSH2 gene, results from a deletion of one nucleotide within intron 14 of the MSH2 gene. This nucleotide position is well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration may weaken the native splice donor site and may result in the creation or strengthening of a novel splice donor site; however, direct evidence is insufficient at this time (Ambry internal data). Based on the available evidence, the clinical significance of this variant remains unclear.
