Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005591.4(MRE11):c.14A>G (p.Asp5Gly), citing Ambry Variant Classification Scheme 2023: The p.D5G variant (also known as c.14A>G), located in coding exon 1 of the MRE11A gene, results from an A to G substitution at nucleotide position 14. The aspartic acid at codon 5 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
