NM_000245.4(MET):c.79A>G (p.Lys27Glu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K27E variant (also known as c.79A>G), located in coding exon 1 of the MET gene, results from an A to G substitution at nucleotide position 79. The lysine at codon 27 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
