Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_015192.4(PLCB1):c.724G>A (p.Val242Ile), citing Ambry Variant Classification Scheme 2023: The p.V242I variant (also known as c.724G>A), located in coding exon 9 of the PLCB1 gene, results from a G to A substitution at nucleotide position 724. The valine at codon 242 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
