Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001370259.2(MEN1):c.1195A>T (p.Ser399Cys), citing Ambry Variant Classification Scheme 2023: The p.S399C variant (also known as c.1195A>T), located in coding exon 8 of the MEN1 gene, results from an A to T substitution at nucleotide position 1195. The serine at codon 399 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
