This variant is reported with the allele frequency 0.002% in the gnomAD Exomes. This variant causes a frameshift starting with codon Leucine 281, changes this amino acid to Tryptophan residue, and creates a premature Stop codon at position 7 of the new reading frame, denoted p.Leu281TrpfsTer7. This variant is predicted to cause loss of normal protein function through protein truncation. However, loss of function variants have not been previously reported to be disease causing. For these reasons, this variant has been classified as Uncertain Significance. The same variant has been detected in heterozygous state in sibling.
