Pathogenic — the classification assigned by Quest Diagnostics Nichols Institute San Juan Capistrano to GRCh37/hg19 7q34-36.3(chr7:142491993-159119707)x3, citing ACMG/ClinGen CNV Guidelines, 2019. This is a single-copy gain (three copies) of the chr7:142491993-159119707 region (~16.63 Mb) on cytogenetic band 7q34-36.3. Submitter rationale: This gain involves over 50 protein-coding genes, including VIPR2 (OMIM 601970). Heterozygous duplications of VIPR2 have been identified individuals with autism and other phenotypes (Firouzabadi 2017, Kushima 2018, Sajan 2013), and one study reported significant enrichment of VIPR2 duplications in individuals diagnosed with schizophrenia (Vacic 2011). There are no similar copy number gains of this region in the general populations of the Database of Genomic Variants. Thus, based on gene count and current medical literature, this CNV is classified as pathogenic. References: Firouzabadi et al., Mol Neurobiol. 2017 Nov;54(9):7019-7027.
