Likely pathogenic for ketotic hypoglycaemia — the classification assigned by Smedley Team, Phenogenomics Group, Queen Mary University of London to NM_020223.4(FAM20C):c.925_926insGGTG (p.Asn309delinsArgTer), citing ACMG Guidelines, 2015: This variant, resulting in a frameshift in FAM20C (p.Asn309Argfs*2) that is predicted to lead to NMD (PVS1), was detected in a patient recruited to the 100,000 Genomes Project under the ketotic hypoglycaemia specific disease category. In addition, this variant is absent from gnomAD and was classified as PM2 leading to an overall pathogenic classification. FAM20C was identified as a novel disease-gene association for ketotic hypoglycaemia in a gene burden study of the 100,000 Genomes Project.
