Submitter rationale: The observed missense variant c.9806G>Ap.Arg3269Lys in ANK2 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The p.Arg3269Lys variant is reported with 0.001% allele frequency in gnomAD Exomes. The amino acid Arg at position 3269 is changed to a Lys changing protein sequence and it might alter its composition and physico-chemical properties. Multiple lines of computational evidence SIFT-Tolerated and Mutation Taster-polymorphism predict no damaging effect on protein structure and function for this variant. The reference amino acid p.Arg3269Lys in ANK2 is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. For these reasons, this variant has been classified as Uncertain Significance.
