Uncertain significance for Donnai-Barrow syndrome — the classification assigned by Neuberg Centre For Genomic Medicine, NCGM to NM_004525.3(LRP2):c.9623A>G (p.Tyr3208Cys), citing ACMG Guidelines, 2015: The observed missense variant c.9623A>Gp.Tyr3208Cys in the LRP2 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is absent in the gnomAD Exomes. The amino acid Tyr at position 3208 is changed to a Cys changing protein sequence and it might alter its composition and physico- chemical properties. Multiple lines of computational evidence Polyphen - Probably damaging and MutationTaster - Disease causing predict a damaging effect on protein structure and function for this variant. The residue is conserved by GERP++ and PhyloP across 100 vertebrates. For these reasons, this variant has been classified as Uncertain Significance. A different missense variant in the same gene has been detected in the spouse.
