Uncertain significance for Abnormality of the nervous system; Autosomal recessive ataxia, Beauce type — the classification assigned by Neuberg Centre For Genomic Medicine, NCGM to NM_182961.4(SYNE1):c.20132G>T (p.Ser6711Ile), citing ACMG Guidelines, 2015: The observed missense variant c.20132G>Tp.Ser6711Ile in the SYNE1 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is absent in the gnomAD Exomes. The amino acid Ser at position 6711 is changed to a Ile changing protein sequence and it might alter its composition and physico-chemical properties. Multiple lines of computational evidence Polyphen - Damaging, SIFT - Damaging and MutationTaster - Disease causing predict a damaging effect on protein structure and function for this variant. The residue is conserved by GERP++ and PhyloP across 100 vertebrates. For these reasons, this variant has been classified as Uncertain Significance.
