NM_032387.5(WNK4):c.2263A>G (p.Met755Val) was classified as Uncertain significance for Abnormality of the kidney; Pseudohypoaldosteronism type 2B by Neuberg Centre For Genomic Medicine, NCGM, citing ACMG Guidelines, 2015. This variant lies in the WNK4 gene (transcript NM_032387.5) at coding-DNA position 2263, where A is replaced by G; at the protein level this means replaces methionine at residue 755 with valine — a missense variant. Submitter rationale: The missense variant c.2263A>G p.Met755Val in the WNK4 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The variant is absent in the gnomAD. The amino acid Met at position 755 is changed to a Val changing protein sequence and it might alter its composition and physico-chemical properties. Computational evidence Polyphen, SIFT and MutationTaster predicts conflicting evidence on protein structure and function for this variant. The amino acid change p.Met755Val in WNK4 is predicted as conserved by PhyloP across 100 vertebrates. For these reasons, this variant has been classified as Uncertain Significance.
