NM_000540.3(RYR1):c.5632G>A (p.Glu1878Lys) was classified as Uncertain Significance for Malignant hyperthermia, susceptibility to, 1 by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015. This variant lies in the RYR1 gene (transcript NM_000540.3) at coding-DNA position 5632, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 1878 with lysine — a missense variant. Submitter rationale: This missense variant replaces glutamic acid with lysine at codon 1878 of the RYR1 protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with anesthetic-associated rhabdomyolysis (PMID: 36280855). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
