NM_000053.4(ATP7B):c.3413-1G>C was classified as Uncertain Significance for Wilson disease by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015. This variant lies in the ATP7B gene (transcript NM_000053.4) at the canonical splice acceptor site of the intron immediately before coding-DNA position 3413, where G is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This variant causes a c.G>C nucleotide substitution at the -1 position of intron 15 of the ATP7B gene. Splice site prediction tools suggest that this variant may have a significant impact on RNA splicing. To our knowledge, this prediction has not been investigated in published RNA studies. This variant has not been reported in individuals affected with ATP7B-related conditions in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Due to insufficient evidence, this variant is classified as Variant of Uncertain Significance.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
