This variant lies in the PKP2 gene (transcript NM_001005242.3) at coding-DNA position 1976, where T is replaced by C; at the protein level this means replaces leucine at residue 659 with serine — a missense variant. Submitter rationale: This missense variant replaces leucine with serine at codon 703 of the PKP2 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been observed in compound heterozygous state with a pathogenic PKP2 variant in an individual affected with early-onset arrhythmogenic cardiomyopathy with severe heart failure (PMID: 30830208). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
