Uncertain Significance for Malignant hyperthermia, susceptibility to, 5 — the classification assigned by All of Us Research Program, National Institutes of Health to NM_000069.3(CACNA1S):c.2361-44_2361-6del, citing ACMG Guidelines, 2015. This variant lies in the CACNA1S gene (transcript NM_000069.3) at 44 bases into the intron immediately before coding-DNA position 2361 through 6 bases into the intron immediately before coding-DNA position 2361, deleting this region. Submitter rationale: This variant causes a deletion of 39 nucleotides in intron 17 splice acceptor site of the CACNA1S gene. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with CACNA1S-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
